Pitch control is easier with larger targets. Try pitching the glass at a fireplace or other large object; it'll break quite easily.

If you put a straw in it, you'll have an easier time finding the right pitch. The correct pitch will wiggle the straw

The hard part is singing it. Finding and keeping that frequency at a decent enough amplitude to transfer sufficient energy into the glass.

Yes, it matters. The trick is to reach a note that causes resonance, which you will know you've reached when the glass vibrates a lot.
Wouldn't holding the glass with your hand dampen the vibration? A solid hard holder would help, no?
I would assume that the intensity of the resonant vibration is great enough that the dampening from a hand is irrelevant.
The vibrating part is the glass "bowl" or cylinder. The stem has very little effect on it when held.
