The Joker's joke or the meaning behind the whole situation?
And the joke simply showed that the second guy, who said "do you think I'm crazy?", doesn't realize that he can't walk the lightbeam at all
Mainly the jokers joke, but the meaning would be nice too.
The Joker put Batman through shit to show him what "one bad day can do to you". In the end the Joker gets him to laugh, thus "breaking" him

Some interpret it as Batman breaking the joker and killing him, the writers have stated otherwise, but still it's too damning not to.

I thought Alan Moore said he was killing the Joker, but the artist Brian Bolland said he wasn't.
